چکیده

The regulation of the intermediary steps of the catecholamine-stimulated GTPase cycle by &adrenergic agonists and Mg2+ was investigated using unilamellar phosphatidylethanolaminephosphatidylserine vesicles that contained purified &adrenergic receptor and the stimulatory GTP-binding protein of the adenylate cyclase system, G. The steady-state turnover number of the agonist-stimulated GTPase, normalized according to the receptor-responsive pool of G., was 0.8 min-' for untreated vesicles and 1.7 min" for vesicles that had been treated with dithiothreitol to activate the receptors. The binding and release of [~Y-"~P]GTP, ['HI GTP, and [y32P]GTP were used to measure the binding and hydrolysis of GTP and the release of GDP. Agonist-liganded receptor stimulated both the binding of GTP and the release of the GDP product, and GDP release per se did not appear to be the mechanism by which receptor stimulated the binding of GTP. Both processes displayed apparent first order rate constants of about 0.5 min" for untreated vesicles and both rates increased about 5-fold after dithiothreitol treatment. Both processes were formally catalytic with respect to receptor, in that several (up to 8) molecules of G. were stimulated per molecule of receptor. The hydrolysis of G.-GTP to G.-GDP was unaltered by agonist and occurred with a rate constant of about 4 min-l. The rates of these partial reactions were consistent with the overall rate of steady-state hydrolysis and with the ability of the agonist-liganded receptor to promote the formation of sufficient G.-GTP to fully stimulate adenylate cyclase in a native membrane. The Mg2+ dependence of agonist-stimulated, steadystate GTPase activity appeared to consist of at least two, distinct Mg2+-requiring processes. Very low concentrations of Mg2+ (-20 nM) were required for hydrolysis of G.*GTP, and 10 MM Mg2+ was required to maximize the initial rate of agonist-stimulated [cY-'~P] GTP binding.

Enhancement of the dense-core vesicle secretory cycle by glucocorticoid differentiation of PC12 cells: characteristics of rapid exocytosis and endocytosis.

The secretory cycle of dense-core vesicles (DCVs) in physiologically stimulated patch-clamped PC12 cells was analyzed using both amperometry and capacitance measurements. Untreated cells had low or undetectable Ca currents and sparse secretory responses to short depolarizations. Dexamethasone (5 microM) treatment for 5-7 d tripled Ca current magnitude and dramatically increased quantal secretio...

Pseudomonas aeruginosa ExoT is a Rho GTPase-activating protein.

Transient intracellular expression of ExoT in CHO cells stimulated cell rounding and actin reorganization. Biochemical studies showed that ExoT was a GTPase-activating protein for RhoA, Rac1, and Cdc42. Together, these data show that ExoT interferes with Rho signal transduction pathways, which regulate actin organization, exocytosis, cell cycle progression, and phagocytosis.

Exocytosis in chromaffin cells. Possible involvement of the heterotrimeric GTP-binding protein G(o).

The use of non-hydrolyzable analogues of GTP in permeabilized secretory cells suggests that guanine nucleotide-binding regulatory proteins (G proteins) may be involved in regulated exocytosis. Because GTP analogues are known to modulate both monomeric low molecular mass G proteins and heterotrimeric G proteins, we have examined the effect of mastoparan, an activator of heterotrimeric G proteins...

Overexpression of RAN1 in rice and Arabidopsis alters primordial meristem, mitotic progress, and sensitivity to auxin.

Ran is an evolutionarily conserved eukaryotic GTPase. We previously identified a cDNA of TaRAN1, a novel Ran GTPase homologous gene in wheat (Triticum aestivum) and demonstrated that TaRAN1 is associated with regulation of genome integrity and cell division in yeast (Saccharomyces cerevisiae) systems. However, much less is known about the function of RAN in plant development. To analyze the pos...

Reconstitution of Catecholamine-Stimulated Guanosinetriphosphatase Activity t

&Adrenergic receptors were partially purified from turkey erythrocyte membranes by alpr.enolo1-agarose chromatography to 0.25-2 nmol/mg of protein, and the stimulatory guanosine 5’-triphosphate (GTP) binding protein of adenylate cyclase (G,) was purified from rabbit liver. These proteins were reconstituted into phospholipid vesicles by addition of phospholipids and removal of detergent by gel f...
